Adaptive behavior of Scotocryptus melitophilus Reitter (Coleoptera, Leiodidae) to live with its host Melipona quadrifasciata Lepeletier (Hymenoptera, Apidae) Zoologia
Bezerra,José Maurício Dias; Peruquetti,Rui Carlos; Kerr,Warwick Estevam.
Forty six beetles Scotocryptus melitophilus Reitter, 1881 collected in a nest of Melipona quadrifasciata Lepeletier, 1836 were studied. All beetles were put together in a large petri dish with 14 males, 14 virgin queens and 14 workers of M. quadrifasciata, which was not aggressive towards the S. melitophilus and flew with 3 to 10 beetles attached to them. When groups of 20 workers, 20 males and 20 virgin queens were put separately in three petri dishes, the workers become attractive to beetles after they were seven days old, while the males after the 19 days old. Only one virgin queen five days old attracted one beetle. The anatomic parts of the workers bees more attractive to the beetles when smeared to a filter paper were the head (32%), abdomen (29%)...

Antimicrobial activity of honeys from two stingless honeybee species and Apis mellifera (Hymenoptera: Apidae) against pathogenic microorganisms Acta Amazonica
Cruz,Carolinie Batista Nobre da; Pieri,Fabio Alessandro; Carvalho-Zilse,Gislene Almeida; Orlandi,Patrícia Puccinelli; Nunes-Silva,Carlos Gustavo; Leomil,Luciana.
Honeys are described possessing different properties including antimicrobial. Many studies have presented this activity of honeys produced by Apis mellifera bees, however studies including activities of stingless bees honeys are scarce. The aim of this study was to compare the antimicrobial activity of honeys collected in the Amazonas State from Melipona compressipes, Melipona seminigra and Apis mellifera against Staphylococcus aureus, Enterococcus faecalis, Escherichia coli, Chromobacterium violaceum, and Candida albicans. Minimum inhibitory concentrations were determined using the agar dilution method with Müller-Hinton agar (for bacteria) or Saboraud agar (for yeast). Staphylococcus aureus and E. faecalis were inhibited by all honeys at concentrations...

Comparative study on the use of specific and heterologous microsatellite primers in the stingless bees Melipona rufiventris and M. mondury (Hymenoptera, Apidae) Genet. Mol. Biol.
Lopes,Denilce Meneses; Campos,Lúcio Antônio de Oliveira; Salomão,Tânia Maria Fernandes; Tavares,Mara Garcia.
Due to their high degree of polymorphism, microsatellites are considered useful tools for studying population genetics. Nevertheless, studies of genetic diversity in stingless bees by means of these primers have revealed a low level of polymorphism, possibly the consequence of the heterologous primers used, since in most cases these were not specifically designed for the species under consideration. Herein we compared the number of polymorphic loci and alleles per locus, as well as observed heterozygosity in Melipona rufiventris and M. mondury populations, using specific and heterologous primers. The use of specific primers placed in evidence the greater frequency of polymorphic loci and alleles per locus, besides an expressive increase in observed...

Cytogenetic characterization of Melipona rufiventris Lepeletier 1836 and Melipona mondury Smith 1863 (Hymenoptera, Apidae) by C banding and fluorochromes staining Genet. Mol. Biol.
Lopes,Denilce Meneses; Pompolo,Sílvia das Graças; Campos,Lúcio Antônio de Oliveira; Tavares,Mara Garcia.
The stingless bees Melipona rufiventris and M. mondury were analyzed cytogenetically by conventional staining with Giemsa, C-banding and sequential staining with the fluorochromes CMA3/DA/DAPI. Both species presented 2n = 18 and n = 9, except for one colony of M. rufiventris, in which some individuals had 2n = 19 due to the presence of a B chromosome. After Giemsa staining and C-banding the chromosomes appeared very condensed and presented a high heterochromatic content, making it difficult to localize the centromere and therefore to visualize the chromosomes morphology. The constitutive heterochromatin was located in interstitial chromosome regions covering most of the chromosomes extension and consisted mainly of AT, as shown by DAPI staining. The...

Genetic breeding on the bee Melipona scutellaris (Apidae, Meliponinae) Acta Amazonica
Barros,José de Ribamar Silva.
A selection of queens of Melipona scutellaris through the most productive colonies were carried out during eight months in an orange honeyflow. Each of the colonies was evaluated by its production, that is, the gross weight production ( pollen, brood, geopropolis and wax of each hive). With this data a coefficient of repeatability was estimated by the intraclass correlation method, obtained r = 0.835 ± 0.071. The repeatibility is very high showing that the analysed data (production) is repeatable. Selection was then carried out using the regression coefficient of each colony and the respective production gain. Using these data the colonies were divided into three groups according to the method Vencovsky and Kerr (1982): a with the colonies of highest...

Honey physicochemical properties of three species of the brazilian Melipona Anais da ABC (AABC)
Lage,Lorena G.A.; Coelho,Lívia L.; Resende,Helder C.; Tavares,Mara G.; Campos,Lucio A.O.; Fernandes-Salomão,Tânia M..
Physicochemical analyses were carried out to evaluate 27 samples of honeys from three species of the Brazilian genus Melipona (M. capixaba, M. rufiventris and M. mondury) from Espírito Santo and Minas Gerais States. The parameters water activity (Aw), percentage of soluble solids (Brix %), pH, acidity (meq/Kg) and moisture (%) were evaluated. The honey characteristics obtained from these samples were very similar to the ones from other Melipona species. However, regarding the honey from Apis (honey bee), only the pH values were similar. The low pH value and the high acidity detected in Melipona honey are potential factors for increasing the honey shelf life because they do not provide favorable conditions for the microbial development. On the other hand,...

Insights into the Melipona scutellaris (Hymenoptera, Apidae, Meliponini) fat body transcriptome Genet. Mol. Biol.
Sousa,Cristina Soares de; Serrão,José Eduardo; Bonetti,Ana Maria; Amaral,Isabel Marques Rodrigues; Kerr,Warwick Estevam; Maranhão,Andréa Queiroz; Ueira-Vieira,Carlos.
The insect fat body is a multifunctional organ analogous to the vertebrate liver. The fat body is involved in the metabolism of juvenile hormone, regulation of environmental stress, production of immunity regulator-like proteins in cells and protein storage. However, very little is known about the molecular mechanisms involved in fat body physiology in stingless bees. In this study, we analyzed the transcriptome of the fat body from the stingless bee Melipona scutellaris. In silico analysis of a set of cDNA library sequences yielded 1728 expressed sequence tags (ESTs) and 997 high-quality sequences that were assembled into 29 contigs and 117 singlets. The BLAST X tool showed that 86% of the ESTs shared similarity with Apis mellifera (honeybee) genes. The...

Prediction of social structure and genetic relatedness in colonies of the facultative polygynous stingless bee Melipona bicolor (Hymenoptera, Apidae) Genet. Mol. Biol.
Reis,Evelyze Pinheiro dos; Campos,Lucio Antonio de Oliveira; Tavares,Mara Garcia.
Stingless bee colonies typically consist of one single-mated mother queen and her worker offspring. The stingless bee Melipona bicolor (Hymenoptera: Apidae) shows facultative polygyny, which makes this species particularly suitable for testing theoretical expectations concerning social behavior. In this study, we investigated the social structure and genetic relatedness among workers from eight natural and six manipulated colonies of M. bicolor over a period of one year. The populations of M. bicolor contained monogynous and polygynous colonies. The estimated genetic relatedness among workers from monogynous and polygynous colonies was 0.75 ± 0.12 and 0.53 ± 0.16 (mean ± SEM), respectively. Although the parental genotypes had significant effects on genetic...

Removal of clay by stingless bees: load size and moisture selection Anais da ABC (AABC)
COSTA-PEREIRA,RAUL.
Some organisms disperse energy, associated with the transportation of resource, which is not necessarily food. Stingless bees of Central Amazonia (Melipona flavolineata and M. lateralis) collect clay in banks along streams for nest building. The moisture of the clay varies along the bank, and bees collect clay from specific location, indicating that there is some sort of preference regarding their selection. This study aims at identifying: if larger bees carry more clay; if there is a preference for moisture of substrates; and if bees are less efficient accumulating and transporting clay when it is wet. In order to do so, I measured the size of the bees and of the pellets of clay found in the corbicula. I set up a field experiment to test substrate...

Variation and genetic structure of Melipona quadrifasciata Lepeletier (Hymenoptera, Apidae) populations based on ISSR pattern Genet. Mol. Biol.
Nascimento,Marcília A.; Batalha-Filho,Henrique; Waldschmidt,Ana M.; Tavares,Mara G.; Campos,Lucio A.O.; Salomão,Tânia M.F..
For a study of diversity and genetic structuring in Melipona quadrifasciata, 61 colonies were collected in eight locations in the state of Minas Gerais, Brazil. By means of PCR analysis, 119 ISSR bands were obtained, 80 (68%) being polymorphic. He and H B were 0.20 and 0.16, respectively. Two large groups were obtained by the UPGMA method, one formed by individuals from Januária, Urucuia, Rio Vermelho and Caeté and the other by individuals from São João Del Rei, Barbacena, Ressaquinha and Cristiano Otoni. The Φst and θB values were 0.65 and 0.58, respectively, thereby indicating high population structuring. UPGMA grouping did not reveal genetic structuring of M. quadrifasciata in function of the tergite stripe pattern. The significant correlation between...
